GATA-2 is a 546-amino acid protein that is encoded by the GATA2 gene, located on chromosome 3q21 in humans. The protein consists of two highly conserved GATA-type zinc finger domains, which are essential for DNA binding and transcriptional regulation. The N-terminal zinc finger domain (NF1) is involved in DNA recognition, while the C-terminal zinc finger domain (NF2) is required for transcriptional activation.

In rare cases, a patient inherits a GATA-2 mutation, but their hematopoietic stem cells acquire a second , spontaneous mutation that restores function. These patients may have normal blood counts but still suffer from lymphedema (since the endothelium retains the mutation). gata-2
