In a standard blood sample, FSC-A and SSC-A (which measures internal complexity/granularity) can distinguish between different leukocyte types:

FSC-A tells you about size; tells you about granularity/complexity. Never gate on FSC-A alone. Always use a bivariate plot of FSC-A vs. SSC-A . This separates cells by both size (FSC) and internal complexity (SSC). For example, granulocytes have high FSC-A (large) and high SSC-A (granular), while monocytes have medium FSC-A but low SSC-A. In a standard blood sample, FSC-A and SSC-A
